Customs and Excise is leaving its offices next to Douglas Railway Station.
The government says its Customs and Immigration division has operated from two separate buildings since January last year.
However, the office in Custom House on North Quay will shut on Friday, December 12.
The division will then move to the ground floor of Central Government Offices on Bucks Road from Monday, December 15.
