It says it acted after receiving complaints about the vehicles at the store on Lake Road in Douglas.
The DoI is warning owners than unless removed within 14 days, it will tow the cars away.
It's thought a number of them - which include Range Rovers and Chryslers - had been parked in the Lord Street car park, which closed to vehicles three weeks ago.
